Exploring Cannabis sativa L for Anti-Alzheimer Potential: An Extensive Computational Study including Molecular Docking, Molecular Dynamics, and ADMET Assessments

Abstract

Introduction: Cholinesterase enzymes play a pivotal role in hydrolyzing acetylcholine, a neurotransmitter crucial for memory and cognition, into its components, acetic acid, and choline. A primary approach in addressing Alzheimer’s disease symptoms is by inhibiting the action of these enzymes.

Methods: With this context, our study embarked on a mission to pinpoint potential Cholinesterase (ChE) inhibitors using a comprehensive computational methodology. A total of 49 phytoconstituents derived from Cannabis sativa L underwent in silico screening via molecular docking, pharmacokinetic and pharmacotoxicological analysis, to evaluate their ability to inhibit cholinesterase enzymes. Out of these, two specific compounds, namely tetrahydrocannabivarin and Δ-9- tetrahydrocannabinol, belonging to cannabinoids, stood out as prospective therapeutic agents against Alzheimer’s due to their potential as cholinesterase inhibitors. These candidates showcased commendable binding affinities with the cholinesterase enzymes, highlighting their interaction with essential enzymatic residues.

Results: They were predicted to exhibit greater binding affinities than Rivastigmine and Galantamine. Their ADMET assessments further classified them as viable oral pharmaceutical drugs. They are not expected to induce any mutagenic or hepatotoxic effects and cannot produce skin sensitization. In addition, these phytoconstituents are predicted to be BBB permeable and can reach the central nervous system (CNS) and exert their therapeutic effects. To delve deeper, we explored molecular dynamics (MD) simulations to examine the stability of the complex formed between the best candidate (Δ-9-tetrahydrocannabinol) and the target proteins under simulated biological conditions. The MD study affirmed that the ligand-ChE recognition is a spontaneous reaction leading to stable complexes.

Bernal’s unwavering dedication to the Bidens, combined with his ruthless demeanor toward perceived adversaries, made him an unpopular figure within the White House. “He would not be welcome at my funeral,” one aide candidly mentioned in Tapper and Thompson’s book.

Despite Jill’s insistence on being addressed as “Dr. Biden” by others, she allowed Bernal the privilege of first-name familiarity. His open identity as a gay man afforded him a unique level of intimacy with the former First Lady. Bernal offered candid opinions on Jill’s appearance as they crafted her public image, frequently highlighted by outlets like *Vogue*.

Even after Joe Biden’s presidency faced criticism during a key debate, Jill maintained her glamorous persona with Bernal’s help. Following the debate, Jill told *Vogue*, “We will continue to fight,” emphasizing that they wouldn’t let those 90 minutes define the presidency’s four years. This defiant stance came despite growing scrutiny over the administration’s performance.

Now, with the Bidens no longer in the White House, Jill faces increasing pressure to disclose what she knew about her husband’s health. The revelation of Joe Biden’s advanced prostate cancer diagnosis has only intensified calls for transparency. “Frankly, the former first lady should certainly speak up about what she saw in regards to her husband and when she saw it, and what she knew,” said White House press secretary Karoline Leavitt in a May 29 briefing.

Biden’s Doctor Gets Subpoena After Resisting Investigation Into His Personal Connection To Biden Family  

The House Committee on Oversight and Accountability has been trying to have a conversation for almost a year with Dr. Kevin O’Connor, former President Joe Biden’s physician. So far, he has refused to cooperate. In a letter sent on Thursday, the committee demanded, with the power of a subpoena, that he sit for a deposition on June 27.

Despite numerous concerning public moments when Biden seemed too old and infirm to handle the grueling work of the presidency, O’Connor gave Biden a clean bill of health. In February 2024, O’Connor assessed Biden as “a healthy, active, robust 81-year-old male, who remains fit to successfully execute the duties of the Presidency,” noted the letter from committee Chair James Comer.

This assessment is nearly impossible to believe because that same month, Special Counsel Robert Hur unsealed his report to the Department of Justice that found problems with Biden’s handling of classified documents. Hur doubted he could secure a conviction and declined to charge Biden because, according to Hur, “Biden would likely present himself to the jury, as he did during our interview with him, as a sympathetic, well-meaning, elderly man with a poor memory.” Hur went on to say, “It would be difficult to convince a jury that they should convict him — by then a former president well into his eighties — of a serious felony that requires a mental state of willfulness.”

Incredibly, Biden remained president until the end of his term, making few appearances and reportedly signing almost everything with a staff-operated autopen, which is also currently under investigation.

Within months of Biden leaving the White House, his family announced he has advanced prostate cancer. How did O’Connor miss that?

The Oversight Committee is investigating more than the state of Biden’s health. It wants to know if O’Connor was personally motivated to produce favorable reports for his own gain.  

“The committee expressed its interest in whether your financial relationship with the Biden family affected your assessment of former President Biden’s physical and mental fitness to fulfill his duties as President,” Comer’s letter said. “Given your connections with the Biden family, the committee sought to understand if you contributed to an effort to hide former President Biden’s fitness to serve from the American people.”

House Oversight subpoenas Biden doctor Kevin O’Connor in probe into alleged mental decline cover-up

House Oversight Committee Chairman James Comer on Thursday issued a subpoena to former President Joe Biden’s physician Kevin O’Connor, calling for his testimony over the alleged cover-up of the former president’s mental decline.

The legal action instructs the physician to appear for a deposition on June 27. The subpoena comes after O’Connor declined to be interviewed voluntarily. 

“On May 22, 2025, the Committee on Oversight and Government Reform requested that you—because of your role as former Physician to the President for President Joe Biden— appear for a transcribed interview on June 25, 2025, broadly regarding ‘the circumstances surrounding your assessment in February 2024 that former President Biden was ‘a healthy, active, robust 81-year-old male, who remains fit to successfully execute the duties of the Presidency,'” Comer wrote. 

“Given your connections with the Biden family, the Committee sought to understand if you contributed to an effort to hide former President Biden’s fitness to serve from the American people,” he continued. “You refused the Committee’s request. However, to advance the Committee’s oversight and legislative responsibilities and interests, your testimony is critical.”

The subpoena comes as the committee and Justice Department probe the perceived cover-up and Biden’s use of an autopen to sign official documents, including presidential pardons, toward the end of his term.

Biden argued on Wednesday that a probe into his cognitive abilities was a “distraction” by Republicans, and reaffirmed that he was responsible for all decisions at the end of his tenure in the Oval Office.

Biden insists ‘I made the decisions’ as Republicans investigate White House autopen use

Former President Joe Biden doubled down on his use of an autopen on Wednesday, insisting that he was in control of the White House during his term in office.

President Donald Trump ordered an investigation into Biden’s administration, alleging that top officials used autopen signatures to cover up the former president’s cognitive decline.

“I made the decisions about the pardons, executive orders, legislation, and proclamations. Any suggestion that I didn’t is ridiculous and false,” Biden said in a statement.

“This is nothing more than a distraction by Donald Trump and Congressional Republicans who are working to push disastrous legislation that would cut essential programs like Medicaid and raise costs on American families, all to pay for tax breaks for the ultra-wealthy and big corporations,” he added.

Trump called on Attorney General Pam Bondi to open investigations into top Biden officials on Wednesday, arguing they may have conspired to deceive the public about his mental state and exercised presidential authority through use of the autopen.

Here We Go: Trump Administration Launches Investigation into Biden Autopen Scandal – Here Are the Details

The long-awaited investigation into Joe Biden’s autopen scandal has begun and several members of his family should start sweating.

Reuters obtained an email from Ed Martin, the Justice Department’s pardon attorney, Ed Martin, telling staff that he has been ordered to investigate clemency allegedly granted by Biden in the final his time in office to his family members and death row inmates.

Martin wrote in the email that he will investigate whether Biden “was competent and whether others were taking advantage of him through use of AutoPen or other means.”

The email states that Martin will specifically focus on the preemptive pardons Biden issued to six members of his crooked family and the clemency he granted to 37 child r*pists, murderers, and other violent thugs on death row.

Biden’s executive orders apparently will not be covered in the investigation.

As TGP readers know, Biden pardoned five members of his family hours before Trump took the oath of office back in January.

Biden issued preemptive pardons to James B. Biden, Sara Jones Biden, Valerie Biden Owens, John T. Owens, and Francis W. Biden for any nonviolent offenses they may have committed from January 1, 2014, through the date of the pardon.

On December 1, Biden pardoned his ne’er do well son Hunter. The pardon, which covered any federal crimes by Hunter from 2014 to 2024, ended a case that was blown open by IRS whistleblowers who spoke out after the Biden Justice Department slow-walked an investigation into Hunter, allowing the statute of limitations to pass on some charges and offering Hunter a sweetheart plea deal that was ultimately rejected by an incredulous federal judge in Delaware resulting in the appointment of a special counsel by Attorney General Merrick Garland in the case.

News of this investigation comes after Martin identified the gatekeepers involved in the autopen scandal late last month.

Former President Bill Clinton raised some eyebrows this morning after making some unbelievable comments about Joe Biden and health.

As TGP readers know, Biden has been hit with multiple elements of awful news from his prostate cancer diagnosis, the Tapper book, which has unveiled several disturbing details about the former White House occupant’s cognitive state, and the ongoing auto-pen scandal.

On Sunday, CBS News correspondent Tracy Smith interviewed Clinton and decided to ask him about his experiences with Biden. What he had to say in response is insulting to the intelligence of any sentient human being.

“There’s this book that talks about Joe Biden and the people around him, seeing that he had cognitive and physical decline,” Smith said. “Did you ever have a moment with him where you thought maybe he was unfit to run for president?”

“No, I thought he was a good president,” Clinton replied. “The only concern I thought he had to deal with was, could anybody do that job until they were 86?”

“I had never seen him, and walked away thinking he can’t do this anymore. He was always on top of his brief.”

Energy group says Biden had no knowledge of climate change EOs, doubt validity of autopen use

Apro-energy group scrutinized eight of former President Joe Biden’s executive orders which pertained to climate and energy issues, but their research found no evidence that Biden ever spoke publicly about the contents of the climate change-oriented EOs. The group also asserts that the signatures on the EOs match Biden’s autopen signature instead of his genuine signature, thus calling into question whether the president ever knew about the executive orders. 

Power the Future, the organization who examined the orders, is now urging investigations from multiple bodies to determine if Biden knew of the executive orders and, if not, who did, and what course of action should be taken next. 

No evidence Biden knew about the EO’s signed with his name

Daniel Turner, the founder and executive director of Power the Future, spoke to the Furthermore with Amanda Head podcast and said, “The curious thing about these executive orders is that we found no evidence at all that the President spoke of them on the record. He wasn’t asked a question by the media. He wasn’t stopped on Air Force One. He didn’t give a speech about it.”

“There’s no evidence that the president was cognizant that this was done, that he directed it, that he was part of the decision. There was never any follow-up,” Turner continued. “The only evidence we have that the President signed it is the autopen signature and then some little statement on social media.”

Turner said that his organization highlighted these specific orders because of their scope, how much damage they did to the energy industry and, by extension, to the overall economy and national security.

Power the Future sent their findings to multiple federal agencies, including the Department of Justice, the Environmental Protection Agency, the Department of the Interior, the Department of Energy, along with the House and Senate Oversight Committees. 

Among the most critical of Biden’s executive actions on climate and energy include an Inauguration Day executive order in 2021 committing the federal government to net-zero emissions by 2050, a 2023 order banning arctic drilling, and an order requiring “clean energy” artificial intelligence centers, and a last-minute offshore drilling ban shortly before leaving office in 2025.

A Secret Service whistleblower claims that former President Joe Biden was so out of it at the White House that he would “get lost in his closet,” Sen. Josh Hawley revealed Friday.

The stunning level of disorientation is an example of why the Democratic cover-up of Biden’s mental decline is one of the biggest scandals in presidential history, Hawley argued.

“This Secret Service whistleblower actually was assigned to Biden,” the Missouri Republican told Fox News host Sean Hannity.

“He told me that Biden used to get lost in his closet in the mornings at the White House,” Hawley claimed, noting that he spoke to numerous Secret Service agents while investigating the assassination attempts against President Trump.

“I mean, the guy literally stumbling around in the White House residence couldn’t find his way out of his own closet,” Hawley continued. “The president of the United States.

“This is outrageous. We were lied to.”

Hawley’s disclosure of the Secret Service whistleblower’s claim comes amid multiple congressional investigations into the 82-year-old ex-president’s cognitive abilities while in office — and the role his staff played in running the country.

House Oversight Committee Chairman James Comer (R-Ky.) is probing the possible use of an autopen by Biden’s subordinates to sign off on White House directives and pardons without the president’s knowledge. 

Comer has also requested testimony from Dr. Kevin O’Connor, the White House physician during the Biden administration, as part of the probe.

Sleepy Joe Emerges, Declares He Could “Beat The Hell Out Of” Jake Tapper

CNN hypocrite Jake Tapper is paying for his fake about turn on Sleepy Joe Biden’s mental state, with the audience figures for his show plummeting off a cliff.

Tapper has lost a full quarter of his audience since he started aggressively peddling his book documenting Biden’s brain turning to mush, and banging on about his own “humility” after doing a complete 180.

People don’t like inauthenticity, simple as.

It seems that the more he appears on other networks and shows to sell the book, the more audience he loses. The viewing figures are the lowest they’ve been in a decade.

“The Lead with Jake Tapper” averaged 525,000 total viewers from April 28 through May 25, shedding 25% of CNN’s audience in its timeslot from the same period last year in the process, reports Fox News.

Ironically, Tapper claims that he is attempting to redeem himself after failing to report on the Biden shenanigans (he also actively dismissed the notion of Biden’s slipping mental acuity) but has only managed to become more reviled in the process.
